miR RNAi Final Designs

This screen shows the final top-strand, bottom-strand, and double-stranded miR RNAi oligo designs. Note that the antisense sequence to the sequence of interest is used in the final top-strand oligo, to encode the target pre-miRNA. The antisense sequence is shown in green in the strands. The top-strand oligo elements are:

• A 4-nucleotide, 5' overhang (TGCT) complementary to the vector for directional cloning

• A 5' G + the 21-nucleotide antisense sequence derived from the target gene, followed by

• A short spacer of 19 nucleotides to form a terminal loop and

• A short sense target sequence with 2 nucleotides removed to create an internal loop
